基于芯片SELEX得到糖化血红蛋白和总血

糖化血红蛋白能够反映出体内血糖的含量,其对于糖尿病的诊断具有重要意义。本文作者通过制造了一种微流体SELEX芯片,其能够连续并且自动的对包含有磁珠的糖化血红蛋白进行多轮筛选。采用该方法能够减少孵化和分离的时间,同时得到的糖化血红蛋白和总血红蛋白aptamer具有很高的亲和力(解离常数分别为Kd=7.6±3.0nM和7.3±2.2nM)。
AbstractBloodglycatedhemoglobin(HbA1c)levelsreflectingaverageglucoseconcentrationsoverthepastthreemonthsarefundamentalforthediagnosis,monitoring,andriskassessmentofdiabetes.Ithasbeenhypothesizedthataptamers,whicharesingle-strandedDNAsorRNAsthatdemonstratehighaffinitytoalargevarietyofmoleculesrangingfromsmalldrugs,metabolites,orproteins,couldbeusedforthemeasurementofHbA1c.Aptamersareselectedthroughaninvitroprocesscalledsystematicevolutionofligandsbyexponentialenrichment(SELEX),andtheycanbechemicallysynthesizedwithhighreproducibilityatrelativelylowcosts.ThisstudythereforeaimedtoselectHbA1c-andhemoglobin(Hb)-specificsingle-strandedDNAaptamersusinganon-chipSELEXprotocol.AmicrofluidicSELEXchipwasdevelopedtocontinuouslyandautomaticallycarryoutmultipleroundsofSELEXtoscreenspecificaptamersforHbA1candHb.HbA1candHbwerefirstcoatedontomagneticbeads.Followingseveralroundsofselectionandenrichmentwitharandomized40-merDNAlibrary,specificoligonucleotideswereselected.Thebindingspecificityandaffinitywereassessedby北京最好的白癜风医院在那里北京最好看白癜风医院
